Introduction: The main route of transmission of SARS-CoV-2 is through droplets expelled by an infected person, when sneezing, coughing and even talking, and indirectly through fomites.

Objective: Describe the behavior of compliance with health measures for the confrontation of COVID-19 by health personnel in the isolation centers for confirmed cases in the municipality of Sancti Spiritus in the period of 2020-2022.

Methods: A descriptive cross-sectional study was carried out to describe the behavior of compliance with health measures to combat COVID-19 by health personnel in the isolation centers for confirmed cases in the municipality of Sancti Spiritus, Cuba. The sample was made up of 179 health workers who served during the pandemic in isolation centers.

Results: It was observed that the female sex predominated with 63.68% and the age group between 20 to 35 years with 37.43%. Individual protection and general worker protection measures health requirements were met by 88.26% and 80.44% respectively. Conclusions: In the isolation centers of the municipality of Sancti Spiritus, the protocols proposed by the National Directorate of Health of Cuba are applied, with some difficulties due to it being a disease not known until then, sometimes due to exhaustion, the challenge and responsibility of physics, healthcare pressure and in many cases due to lack of knowledge of the activity among non-medical personnel.
